MAST CELL REDISTRIBUTION IN HAMSTER CHEEK PORCH BY TOPICALLY APPLICATED TRIAMCINOLONE ACETONIDE




Abstract
Steroidal ointments possess mainly anti inf lammtory actions and decrease the mast cell number in tissues, but exhibit atrophogenic tendency to the skin and mucosa as a side effect. To know the redistribution of mast cells in hamster cheek pouch by triamcinolone topical application, which is prednisolone derivative, 0. 01 % triamcinolone acetonide paste was applicated to the hamster cheek pouch once a day for 5 weeks and the pouches were observed after 3rd and 5th week each. The .excised pouches from hamsters were fixed with 10 96 neutral buffered formalin, embedded in paraffin, stained with toluidine blue solution, and observed with light microscope.
The results were as follows.
1. The mast cell number was decreased noticeably in 3rd week experimental group, but not in control groups.
2. Necrotic change in 5th week experimental group could be seen, the necrotic tissues were denuded and the mast cells were not present in that area.
3. Adjacent the necrotic center, epithelial thickening and inflammatory changes could be seen, but only a few mast cells were present. In some area, many mast cells were aggregated densely and, also, many small, round mast cells were aggregated around the blood vessel.
